Shiitake: from Japanese Umami to Human Immunity

There are very few foods that are at once an internationally recognized culinary delicacy and an approved drug. Lentinula edodes — Shiitake — is one of them. In Asian cuisine it is the source of deep umami, that fifth taste that makes Japanese broths impossible to imitate without it. In medicine, its polysaccharide Lentinan was the first mushroom-derived immunomodulatory agent approved as an injectable drug in Japan, in the 1980s.

Lentinan: the polysaccharide that activated integrative oncology

Lentinan is a beta-1,3-glucan with beta-1,6 branches, isolated specifically from the fruiting body of Shiitake. Its mechanism of action is striking: rather than killing cancer cells directly, it activates the immune-system cells that do. In the research it has been described to:

  • Increase the activity of macrophages and NK cells.
  • Stimulate the production of interferons and TNF-α (tumor necrosis factor).
  • Inhibit tumor angiogenesis factors — reducing the blood supply to tumors.

Multiple Japanese clinical studies have reported that injectable Lentinan, combined with standard chemotherapy for gastric cancer, was associated with significantly improved survival rates. The oral extract shows more moderate activity, relevant to general well-being.

Eritadenine and cholesterol

Shiitake contains eritadenine, an uncommon compound that alters the metabolism of hepatic phospholipids and accelerates the clearance of circulating cholesterol, associated in the literature with reductions in blood LDL.

The mechanism differs from that of statins — without their adverse effects on the liver and muscle — making it a complement of interest for people with borderline cholesterol or those seeking natural support.

Other documented benefits

  • Antiviral: Lentinan and other Shiitake polysaccharides show inhibitory activity against several viruses, including HIV and the influenza virus.
  • Antibacterial: Shiitake extracts show activity against Staphylococcus aureus, E. coli, and H. pylori.
  • Oral health: A low-molecular-weight fraction of Shiitake extract interferes in vitro with the adhesion and biofilm formation of cariogenic and periodontal bacteria such as Streptococcus mutans and Prevotella intermedia, acting on their ability to attach to surfaces rather than on total elimination of the flora; on the basis of this selective mechanism, its authors have proposed the extract as a base for daily oral-hygiene products.

How to use it

  • Fresh or dehydrated: In soups, broths, stir-fries. Heat does not destroy Lentinan; it improves its bioavailability by breaking down the cell walls.
  • Powdered extract: 1–2 g per day for consistent functional support.
  • Culinary notes: Shiitake powder adds umami depth to broths, sauces, and dressings without competing with other flavors.

Precautions

  • Shiitake dermatitis: A small proportion of people (especially with exposure to raw Shiitake) may develop a characteristic rash known as “shiitake dermatitis” or flagellate dermatitis. Cooked, the risk practically disappears.
  • People with autoimmune diseases — medical supervision required.
